In December 1980, Gaddafi reinvaded Chad at the request of the FAP-controlled GUNT government to aid in the civil war; in January 1981, Gaddafi suggested a political merger. The Organisation of African Unity (OAU) rejected this and called for a Libyan withdrawal, which came in November 1981. The civil war resumed, and Libya sent troops back in. In 1982, the GUNT government was overthrown by Habré's forces and Oueddei fled to Libya, where Gaddafi provided him with arms to continue to guerrilla war against Habré. In November 1984, Gaddafi met with French President François Mitterrand; both agreed to withdraw from Chad. Oueddei broke with Gaddafi in 1985 due to the former's intentions to negotiate a truce with Habré. Consequently, he was placed under house arrest by Gaddafi and allegedly arrested by Libyan police and shot in the stomach. Oueddei survived the shooting and fled to Algeria, but continued to claim he and Gaddafi enjoyed a good relationship. When Gaddafi ordered the remnant of GUNT to attack Habré in February 1986 in violation of his agreement with Mitterrand, France launched Operation Épervier, which escalated into the Toyota War.
=== Escherichia coli === A popular system utilized is Escherichia coli because of its rapid growth rate (~20–30 minutes), capacity for continuous fermentation and relatively low cost. Additionally, yeast has the capacity to express a high relative volume of heterologous protein. Specifically, up to 30% of proteins produced in yeast can be the heterologous gene product. There also are safe strains of E. coli that have been successfully generated to scale up production. In addition to E. coli's attractive host properties, this host is incredibly popular due to researchers having a large amount of knowledge about its genetics, including the complete genomic sequence. However, issues arise either due to the sequence of the gene of interest and those that are due to the limitations of E. coli as a host. For example, proteins expressed in large amounts in E.coli tend to precipitate and aggregate, which then requires another denaturation, renaturation recovery method. Finally, E. coli is only optimally effective in specific conditions dependent on the gene being inserted.

=== Laboratory synthesis === The chemical synthesis of DMT has been described. It can be synthesized through several possible pathways from different starting materials. The two most commonly encountered synthetic routes are through the reaction of indole with oxalyl chloride followed by reaction with dimethylamine and reduction of the carbonyl functionalities with lithium aluminium hydride to form DMT. The second commonly encountered route is through the N,N-dimethylation of tryptamine using formaldehyde followed by reduction with sodium cyanoborohydride or sodium triacetoxyborohydride. Sodium borohydride can be used but requires a larger excess of reagents and lower temperatures due to it having a higher selectivity for carbonyl groups as opposed to imines. Procedures using sodium cyanoborohydride and sodium triacetoxyborohydride (presumably created in situ from cyanoborohydride though this may not be the case due to the presence of water or methanol) also result in the creation of cyanated tryptamine and beta-carboline byproducts of unknown toxicity while using sodium borohydride in absence of acid does not. Bufotenine, a plant extract, can also be synthesized into DMT. Alternatively, an excess of methyl iodide or methyl p-toluenesulfonate and sodium carbonate can be used to over-methylate tryptamine, resulting in the creation of a quaternary ammonium salt, which is then dequaternized (demethylated) in ethanolamine to yield DMT. The same two-step procedure is used to synthesize other N,N-dimethylated compounds, such as 5-MeO-DMT.

== Amino-acid sequences == Analysis of the suite of known cyclotides reveals many sequence similarities that are important for understanding their unique physico-chemical properties, bioactivities and homology. The cyclotides fall into two main structural subfamilies. Moebius cyclotides, the less common of the two, contain a cis-proline in loop 5 that induces a local 180° backbone twist (hence likening it to a Möbius strip), whereas bracelet cyclotides do not. There is smaller variation in sequences within these subfamilies than between them. A third subfamily of cyclotides are trypsin inhibitors and are more homologous to a family of non-cyclic trypsin inhibitors from squash plants known as knottins or inhibitor cystine knots than they are to the other cyclotides. It is convenient to discuss sequences in terms of the backbone segments, or loops, between successive cysteine residues. The six cysteine residues are absolutely conserved throughout the cyclotide suite and presumably contribute to preserving the CCK motif. Although the cysteines appear essential to maintaining the overall fold, several other residues highly conserved in cyclotides are thought to provide additional stability. Throughout the known cyclotides loop 1 is the most conserved. Apart from the six cysteine residues, the glutamic acid and serine/threonine residues of loop 1 are the only residues to have 100% identity across the bracelet and Möbius subfamilies. Furthermore, the remaining residue of this loop exhibits only a conservative change i.e. glycine/alanine.
